Serangkaian kebijakan diberlakukan mulai dari pengosongan dukuh secara paksa hingga terciptalah dukuh baru bernama Dukuh Ngradinan serta tuntutan penyelesaian panen hanya dalam waktu dua minggu. Dua tahun lamanya menempati dusun baru dan belum habis penderitaan yang mereka alami, kemudian timbullah masalah baru berupa kekeringan yang memengaruhi hasil panen para penduduk. Segala upaya dilakukan untuk mengatasi hal tersebut, namun hasilnya nihil. Banyak penduduk dukuh akhirnya memutuskan berpindah ke desa lain untuk bertahan hidup.
Namun, lain halnya dengan Sesepuh. Sosok tetua yang amat dihormati di dukuh itu, memutuskan bertahan lantaran tak ingin meninggalkan ladang kesayangannya yang telah dipenuhi singgang padi, satu-satunya harta berharga yang telah ia rawat sepenuh hati. Banyak lika-liku berat yang ia lalui untuk bertahan hidup seorang diri di dukuh yang menjelma seperti desa mati itu sembari berjuang merawat singgang saat kekurangan pasokan air akibat kekeringan. Ia bertahan hidup dengan ditemani Bugel, anjing difabel kesayangannya.
Novel Sesepuh dan Sebuah Dukuh ini menyajikan gambaran mengenai perjuangan antara hidup dan mati yang dilalui Sesepuh di tengah-tengah kerasnya hidup di bawah tekanan kolonialisme Jepang. Bertahan hidup seorang diri ketika dukuhnya menjadi desa mati tak berpenghuni akibat kekeringan yang melanda. Melihat bagaimana ia menghadapi berbagai macam ujian dalam mempertahankan apa yang tersisa dan berharga baginya sekalipun nyawa menjadi taruhannya.
